Lipid analysis of freshwater microalgae: A method study

Abstract

A simple and sensitive method for measuring total lipids in algae is desirable because the biomass available for analysis is often low in growth experiments and in natural waters. Several modifications described earlier of a spectrophotometric method based on the sulphophosphovanilline complex were tested; e.g., the extraction step, type of solvent, and separation of the extract. A verification of the final procedure was made by evaluating a proper standard, following the kinetics of the reaction, and conducting an analytical quality control. The accuracy of the method was verified by comparing the results with the sum of fatty acids, analyzed by means of a Hewlett-Packard GLC-system.
