Abstract

The distribution, process of division and migration of nuclei during conidiogenesis, conidium maturation, conidium germination and germ tube elongation in Macrophomina phaseolina (Tassi) Goid. isolates were studied by the HCl-Giemsa technique. Individual conidiogenous cells were initially uninucleate and the nucleus divided once either immediately before or during conidium initiation. The nuclear division was followed by the migration of one daughter nucleus into the conidial primordium and the formation of a cross-septum at the base. Subsequently all nuclei formed within mature conidia were derived from a single migratory nucleus. Conidia on germination produced germ tubes and the nuclei from the conidia streamed into these germ tubes, often dividing during migration. Generally all the nuclei from the conidia migrated into the germ tubes leaving the conidium enucleate but occasionally one or more nuclei remained in the early stages of conidial germination. The distribution of nuclei and the process of division were also studied in hyphae. Mature, hyaline and pigmented hyphal cells were commonly uninucleate but young growing hyaline, hyphal apices were usually multinucleate. During somatic mitosis structures which could be construed as nucleolus, chromosomes and spindle apparatus complete with centrioles were evident. The commonest chromosome number was 6 but aneuploid and haploid nuclei were also observed.

Kurzfassung

Verbreitung, Teilungsverlauf und Auswanderung der Zellkerne während der Konidienentstehung, Konidienreife und Konidienkeimung sowie Keimschlauchverlängerung von Macrophomina phaseolina (Tassi) Goid. Isolaten wurde mit dem HCl-Giemsa-Verfahren untersucht. Zuerst waren die einzelnen Konidienzellen einkernig, Kernteilung fand statt entweder vor oder während der Konidienentwicklung. Nach der Teilung wanderten die Tochterkerne in das Konidienprimordium aus, und die Querscheidewand entwickelte sich auf der Basis. Alle Kerne, die sich dann in den reifen Konidien entwickelten, stammten von einzelnen ausgewanderten Kernen. Konidien an den durch Keimung erzeugten Keimschläuchen, und die Kerne der in die Keimschläuche strömenden Konidien, teilten sich oft während der Wanderung. Im allgemeinen wanderten alle Kerne in die Keimschläuche ab, so daß sich keine Kerne mehr im Konidium befanden; nur manchmal blieb einer oder mehrere sich im frühen Konidienkeimstadium befindenden Kerne zurück. Verbreitung der Kerne im Verlauf der Teilung in Hpyhen wurden ebenfalls untersucht. Reife, durchsichtige und pigmentierte Hyphenzellen waren gewöhnlich einkernig, aber junge, wachsende, durchsichtige Hyphenspitzen besaßen mehrere Kerne. Einzelne geschwollene Hyphenzellen waren gewöhnlich mehrzellig. Während der somatischen Mitose wurden Strukturen wahrgenommen, die den Kernkörperchen, Kernspindeln, Zentriolen und Chromosomen entsprachen. Die gewöhnliche Zahl der Chromosome war 6, aber aneuploide und haploide Kerne konnten auch beobachtet werden.
